Is that means Truth had knew about the ancient unity between San 'Shyuum and Humanity? And Humanity still no clue about their “forefather” aka the ancient humanity and their actions? How great they were?

The Prophet of Truth (Ord Casto) could of been talking about how Prehistoric Humans, and in-directly the San 'Shyuum, who took it upon themselves to control the shaping sickness (the Flood) before it got out of control, their plan ultimately failed however due to Forerunner interference.
At the current time of development, Bungie planned for the Forerunners to actually be ancient Humans… loosely using their original inspiration from the novel Ringworld, by Larry Niven… now however, 343 Industries have re-affirmed that Forerunners and Prehistoric Humans are different species, birthed in the image of the Precursors, who share similarities.
Contact Harvest establishes that the future Prophets of Truth, Mercy, and Regret wrongly concluded that humans are Forerunners who survived the firing of Halo.

Actually the Halo 3 terminals confirm that Humans and forerunners coexisted.

By Halo 3, the Forerunners and Humanity were different species. This is established in the terminals, where the Librarian references a special species found on Earth.
But Truth did believe they were the same. Another misinterpretation of the Reclaimer concept and what Mendicant Bias told them way back at Harvest.
This quote is one of the most underrated in all of Halo. Not only does it perfectly illustrate the covenant’s point of view in regards to Halo, but it also shows just how deeply the prophet of truth believes in the halo rings and their “salvation”. Truth, along with the other prophets, came to the conclusion that humans are descendants of the forerunners in some capacity(explaining why he needs johnson to light the rings). Truth then says that johnson’s(humanity’s) forefathers set aside their compassion for any other goal other than the supposed “great journey”. Then the quote goes,”I see now why they left you behind. You were weak, and gods, must be strong.”
I love this cutscene for so many reasons, but I think those lines from the prophet of truth are crucial to understanding the rest of the story as a whole. In halo 2, it is surmised that the prophets are using the covenant for their own gain and have completely lied about the great journey or any other religious prospect. But after playing through Halo 3, you realize that they truly believe in the great journey, and all the hardships they’ve thrust upon themselves and the subservient races of the covenant have been for the sake of the great journey. There were never any lies on the prophets’ part. They did everything they did for the sake of their belief, no matter how wrong it truly was.
That’s the beauty of the original story. That’s the true realism of the original games. Every single faction, covenant, human, and flood, are all vying for their own goals, and their own survival. No matter how different their goals may seem, one thing is for certain; the outcome of their conflict will determine what remains of the universe. This scene gives the covenant a new perspective from which to view them. Zealots, through and through.
