Here is a interesting thought. Randall Aiken’s daughter Natalie Aiken is a Spartan 2.1
I will give a little back-story to my thoughts.
In ILB’s there is something called a Spartan 1.1’s. These Spartans were the children of Spartan 1’s(Orion project)
While not better than Spartan 1’s themselfs, they were better than the average human.
So this puts my thoughts that Natalie Aiken is the first “Known” Spartan 2.1. Cause she is the child of a Spartan 2.

I suppose you could use that term for kids of Spartan-IIs, but I don’t really see why it matters. Technically Natalie may have inherited a portion of her father’s genes that might make her physically and/or mentally superior to her peers, but otherwise she is still a perfectly normal little girl. She might have some of the genetic markers that designated her father’s induction into the Spartan Project, but since the UNSC isn’t doing that anymore there really shouldn’t be any issue there.

Granted the Spartan 1.1’s lived a normal life till they were 6. That’s when the more obvious abilities started showing.

And at around age 6 the Spartan-IIs were also shown as having naturally heightened physical and mental abilities as well. But either way this doesn’t much matter for Natalie, as current canon puts her at around 12 years old and she’s not set to be forcefully inducted into any super-soldier program. Based on what we’ve seen Randall has raised her just like any parent would a normal child. We don’t know for sure if or how much of her father’s physical and mental predispositions she’s inherited either. I suppose my point was that, while you can call children of S-IIs "Spartan 2.1"s if you want, it doesn’t really mean anything if they’re living completely normal lives and may or may not end up inheriting any of their parent’s genetic “superiority”.

Still regardless. The term may be useless depending on the situation of Natalie at the end of Nightfall anyway. I will leave this spoiler free for anyone who hasnt seen ep 1 yet.
