The Flood made landfall where Buck and the rest of the ODST squad were last seen. Of course it may have been weeks apart but the Covenant only landed in New Mombassa and presumably the ODST will still fight the Covenant after Halo: ODST.
If the Flood does make an appearance then this could be the chance to make them an enemy that cant be stopped by conventional means. The player would end up viewing the Flood in a different way and constantly checking the corners of every room and street.
i don’t think the Flood should take up the whole game, but the idea of the player being a normal human that could be taken down by one Flood form would be interesting. Since ODST do not have the armor and shields of Spartans.
ODST was a massive missed opportunity. It was billed as a more human experience, no more running around as an invincible supersoldier, but Bungie lost their nerve and turned it into just another Halo - yeah, call shielding “stamina” all you like, you’re not fooling anybody. I’d definitely be down for an ODST sequel that actually had us playing as regular humans. No regenerating health, no mowing through endless waves of bad guys. Make it more focused on stealth and exploration and story and carefully planning your way through the fights you can’t avoid.
I wouldn’t set it in New Mombassa, though. It’d be much more interesting to explore an outer colony world, see how the new multi-species society is taking shape. Almost like an Assassin’s Creed-style game set in the Halo universe.
I feel like ODST was a wasted opportunity for another reason too - it was too focused on shoddy one-liners and an undeveloped love story and has silly, cliched characters.
i.e the stoic commander, witty underling (Romeo), devout Christian ‘All-American’ guy (Dutch) etc.
I had high hopes that ODST would focus more on the human condition, so to speak. Watching the ODST live action trailer (which was amazing), it didn’t focus on that ‘bro’ action, Michael Bay-esque explosions and guns and mowing down enemies. It showed the horrors of war. How insignificant humans were to their enemies.
I love Halo for what it is, that as a Spartan you can mow down your enemies and destroy everything in your way, but Bungie had a chance to show the Halo universe in a different light and it was missed.
If there ever were to be a second ODST, I hope it is a different take on the Halo universe.
I could see this happening in a lot of ways; the second game could take place during Halo 3, or anywhere else around Halo: CE and Halo 2 (again), with new Characters, etc.

Sorry guy but the whole lore story with Halo is about the interaction between flood, the forerunners, humans, and covies. They will return.
OT: I think if 343 made an ODST 2 that had flood, it could possibly be more interesting considering that 343 have clearly stated they want to keep sprint consistent across the game. It’d be the Mona Lisa of Halo games.
